Genomic Profiling of Virulence in the Soybean Cyst Nematode Using Single-Nematode Sequencing

Soybean cyst nematode (SCN) is one of the most important diseases in soybean. Currently, main management strategy relies on planting resistant cultivars. However, overuse a single resistance source has led to selection virulent SCN populations, although mechanisms by which overcomes genes remain unknown. In this study, we used nematode-adapted single-cell RNA-seq approach identify potentially involved breakdown Peking and PI 88788 parental soybean lines. We established for first time full transcriptome individuals allowing us list putative virulence against both major sources. Our analysis identified 48 differentially expressed effectors (secreted proteins required infection) alongside 40 showing evidence novel structural variants, 11 effector containing phenotype-specific sequence polymorphisms. Additionally, differential expression revealed an interesting phenomenon coexpressed gene regions with some effectors. The resulted profoundly altered transcriptome, especially known be parasitism. Several polymorphisms were also specific these nematodes could play role acquisition virulence. On other hand, was very similar avirulent ones exception few genes, suggest distinct Peking.
